The T4 dam Methylase recognizes 5´--GATC--3´ sequences and methylates all adenine (N⁶) residues.

Blocking restriction endonuclease cleavage restricted by dam methylation.
Reaction Conditions
- 1x Methylase reaction buffer in the presence of 160 μ M S-adenosylmethionine (SAM). Incubate at 37℃.
Concentration & Storage Condition
- Store at -20℃. 10 unit/㎕ in 50 mM Tris-HCl, pH 7.5, 50 mM KCl, 1 mM DTT, 10 mM EDTA, 200 ㎍/ml BSA, 50% (v/v) glycerol.
Unit Definition
- One unit is defined as the amount of enzyme required to fully protect against Mbo I cleavage of 1 ㎍ of unmethylasted DNA in 1 hr at 37℃.
Heat Inactivation Condition
- 65℃ for 20 min.
